Elise Rundén Pran

Dr. Rundén-Pran er seniorforsker og seksjonsleder for helseeffektlaboratoriet og europeisk registrert (ERT) toksikolog.

Hun har ekspertise innen toksikologi, nanotoksikologi, nanosikkerhet, cellekulturer, 3D-cellulære modeller, nanogentoksikologi, DNA skade, intracellulær celledødssignalisering, mikroskopi (konfokal, TEM), immunhistokjemi, vevskulturer og nevrodegenerasjon.

Hun var del av senter for fremragende forskning (Sff) i molekylær nevrobiologi og nevrodegenerasjon (CMBN, UiO) som PhD-student og postdok.

Elise har erfaring med human risikovurdering og regulatorisk toksikologi. Hun er partnerleder for EuroNanoMed II prosjektene GEMNS og INNOCENT, og NILU prosjektleder for flere prosjekter finansiert av EU og NFR. Hun har utstrakt ekspertise innen GLP, og er utnevnt medlem av nasjonal komité for godkjenning av ERT toksikologer, Vitenskapskomiteen for mat og miljø (VKM), og medlem av fagrådet for toksikologi.

Helseeffektlaboratoriet er sertifisert for god laboratoriepraksis (GLP) for in vitro human toksisitetstesting. Laboratoriet er blant de ledende i Europa innen nanotoksikologi og nanosikkerhet, og fokusområder er gentoksisitet, karsinogenisitet in vitro, avanserte in vitro modeller, regulatoriske aspekter innen nanosikkerhet, high-throughput nanotoksisitetstesting, intelligente teststrategier og adverse outcome pathways (AOPs).

En utfordring i behandling av brystkreftpasienter er høy tilbakefallsfrekvens. Dette prosjektet har fokus på sirkulerende kreftstamceller, som er en viktig årsak til tilbakefall hos pasientene. Målet med dette prosjektet er å utvikle ny medisin for behandling av brystkreft ved hjelp av målrettet terapi ved bruk av nanopartikler.

Behandlingen baserer seg på kjemoterapi i kombinasjon med epigenetikk, genterapi og radioterapi. Tanken er å kombinere alle disse aspektene i et nytt farmaka – COMBOBOMB – basert på nanomedisin. Safe-by-design prinsippet er gjeldende for hele prosjektet, der toksisitetstesting blir utført parallelt med utviklingen av medisinen. Dette gjøres for å bidra til at sluttproduktet er effektivt uten å ha alvorlige bieffekter.

I dette prosjektet er det partnere fra Slovakia (koordinator), Hellas, Spania, Latvia og Norge (http://www.innocent.pixellaris.com/index.html). Toksisitetstestingen gjøres av de norske partnerne i nært samarbeid med de prosjektpartnerne som utvikler nanopartiklene. Prosjektpartnerne har i fellesskap valgt ut fire cellelinjer som er relevante for brystkreft og som skal brukes for testingen, og cellene har blitt dyrket opp for etablering av cellebanker. Standardprosedyrer (SOPer) har blitt utviklet for dyrkning av cellelinjene og for å undersøke celledød og DNA skade (cyto- og gentoksisitetstester) av nanomaterialene. En vanlig utfordring innen nanotoksikologi, er interferens mellom testmetoden og nanomaterialet som skal testes. Derfor er det er viktig å benytte testmetoder som er validert for testing av nanomaterialer.

For testing av celledød (cytotoksisitet), er det utarbeidet SOPer for ulike testmetoder: To såkalte kolorimetriske metoder der cellene farges med fargestoff (Trypan blue og Alamar blue (AB)) og to fargefrie metoder der celledød måles med såkalt elektrisk impedans eller kolonidannelse (colony forming efficiency (CFE)). Tradisjonelle toksisitetstester i laboratoriet utføres under statiske forhold, og reflekterer således ikke det virkelige liv. UiB har derfor utarbeidet et innovativt oppsett med mikrovæskestrøm som skal etterlikne eksponering av brystkreftceller i kroppen. En avansert 3D in vitro modell for brystkreftceller, som bedre etterlikner in vivo situasjonen i kroppen, er under utvikling av NILU, og UiB har bygget en multikompartment chip med mikrovæskestrøm for bedre vekst av brystkreftceller, tumorassoriserte stromale celler og mikroårer.

For gentoksisitetstesting vil en modifisert versjon av comet assay benyttes for å undersøke DNA trådbrudd og spesifikke oksidative DNA baseskader. NILU har modifisert og validert comet assay, samt AB og CFE, for testing av mange prøver samtidig (såkalt high-throughput-målinger) og testet egnethet for bruk til toksisitetstesting av nanomaterialer. NILU har også etablert tester for å undersøke genmutasjoner (Mouse Lymphoma assay og HPRT), og induksjon av kreft (celletransformasjonsassay; in vitro test for karsinogenisitet). Den sistnevnte testen har også blitt modifisert for high-throughput. UiB og NILU har etablert testsystemene, og har testet tilsendte referanse-nanomaterialer av gull (Au) i de aktuelle cellemodellene for validering av testsystemene til bruk for testing av nanopartikler.

Toksisitet av nanomaterialer avhenger av fysiske og kjemiske egenskaper, som for eksempel størrelse. Det er derfor viktig å analysere størrelse og størrelsesfordeling av nanomaterialet i den løsningen som skal brukes for toksisitetstesting. SOP er utarbeidet for karakterisering av størrelse og størrelsesfordeling av nanopartikler i både stamløsning og i cellekulturmediet, ved bruk av nanopartikkel tracking analyse og dynamisk lysspredning (DLS). Stabilitet av liposomformuleringene i ulike dispergeringsmedier ble analysert med DLS.

Det har vært noen utfordringer knyttet til design av nanopartikler som selektivt tas opp i brystkreftceller, og ikke i normale brystceller. UiB har gjort opptaksanalyser av ulike liposomformuleringer (nanobærere) med ulike overflatebehandlinger, for å finne den mest lovende kandidaten for selektivt opptak i kreftceller og dermed målrettet behandling. Den beste nanopartikkel-kandidaten ble valgt ut for videre behandling, og overflaten modifisert for å øke det selektive opptaket i brystkreft stamcellene. Cytotoksisitet av nanobærerne ble testet, og det ble med real-time elektriske impedansmålinger vist at liposomene ikke induserte toksisitet. Videre arbeid med design og testing av nanobærere for selektivt opptak pågår.
